    Zeta’s Lung Cocktail – Dual Stain.

    Zeta’s Lung Cocktail provides dual staining for p40 and Napsin A, a widely used and diagnostically valuable approach in lung cancer pathology, especially for subtyping non–small cell lung carcinomas (NSCLC) into squamous cell carcinoma (SqCC) and adenocarcinoma (ADC). These are the two major histologic subtypes of NSCLC, and they require distinct treatment approaches and carry different prognoses.

    Zeta’s recombinant rabbit antibody cocktail recognizes both P40 and Napsin A, novel diagnostic markers for adenocarcinoma of the digestive system. p63 consists of two major isoforms-TAp63 and DNp63. DNp63 isoform (identified by anti-p40 antibody) contains an alternative transcriptionally inactive domain. The p40 (clone ZR8) antibody recognizes exclusively DNp63 but not TAp63. p40 reacts with most cases of squamous cell carcinomas of various origins, but not with adenocarcinomas. Napsin is a pepsin-like aspartic proteinase, in the A1 clan of the AA clade of proteinases. There are two closely related napsins, napsin A and napsin B. Napsin A is expressed in type II pneumocytes and in lung adenocarcinomas. The high specificity expression of napsin A (brown) in lung adenocarcinomas is useful to distinguish primary lung adenocarcinomas from lung squamous cell carcinoma (P40 positive, red) and adenocarcinomas of other organs.

    Zeta’s Lung Cocktail can help with accurate subtyping of NSCLC where p40+ / Napsin A– supports squamous cell carcinoma; p40– / Napsin A+ supports adenocarcinoma; double negatives may suggest undifferentiated or poorly differentiated NSCLC; and rare double positives may reflect adenosquamous carcinoma or sampling artifact.

    Zeta’s DUAL STAIN products provide more information from limited tissues, by seeing two related markers on the same slide while also allowing pathologists to see cellular localization and interaction within the tumor environment.  Zeta uses one mouse and one rabbit recombinant monoclonal in each cocktail to allow for differential color results.
